I would fly in order to make the most of time at the destination, especially if work related.

seeings it`s an overnighter, toss up between 1 and 2 but if it`s a boring well made freeway travelling at what seems like snails pace for the conditions, (ala hume between melb and woop woop), i`d take option 2, at least towns break up the monotony. PS....bugger flying !.
